Functional NK surrogate biomarkers for inflammatory recurrent pregnancy loss and recurrent implantation failure

Abstract: Problem: Expansion of circulating NK cells has been related to pregnancy complications. This study aims at investigating several surface NK cell markers to identify a baseline inflammatory profile in women with recurrent pregnancy loss (iRPL) and recurrent implantation failure (iRIF).Method of study: Expression of NKp30, TIGIT, NKp46, and DNAM-1 on total peripheral blood NK subsets, regulatory (CD56 bright CD16 neg ), and cytotoxic (CD56 dim CD16 pos/neg ) NK cells was measured. Results: Eighty-three women were… Show more

“…In addition, AHNAK in the PBMCs was indicated to be valuable in the diagnosis of RPL with a high AUC (0.742). As previously reported, certain genes and miRNAs in PBMCs or immune cells have been considered to be diagnostic or predictive biomarkers for immune-related diseases [ 28 , 29 ]. Compared to serum biomarkers, whether biomarkers in PBMCs have advantages in the diagnosis of immune-related conditions requires further investigation.…”

“…18 Given their suggested important role in pregnancy, several studies explored the association between NK cell numbers and reproductive success, though the results between these studies are not fully conclusive yet. 19 Moreover, in women with RPL, differences in peripheral blood NK cell receptor expression have been described for example by Fukui et al 20 Comins et al 21 and Strobel et al 22 who primarily focused on activating receptors, and by Zhu et al, 23 Ntrivalas et al 24 and Emmer et al 25 focusing on CD16 in combination with inhibitory KIRs and NKG2A. The immune checkpoint receptors have not been thoroughly studied in relation to RPL, except for TIM-3.…”

“…Previous studies have focused on the role of NK cells in recurrent spontaneous abortion and RIF. The expression of NKP30 on cytotoxic NK cells (CD56dim CD16pos/neg) significantly increased in RIF (31). High NK cell numbers may be adisadvantage for ovarian reserve or function (32).…”
