Functional Studies of Synthetic Gramicidin Hybrid Ion Channels in CHO Cells

Abstract: The function of a gramicidin hybrid ion channel in living Chinese hamster ovary (CHO) cells was investigated by the patch clamp method. The synthetic ion channel 1 consists of two cyclohexyl ether amino acids that link two mini-gramicidin strands. With 1 at a concentration of 1.0 microM, an increase in the whole-cell membrane conductance was observed after 1.37 min. The conductance showed larger currents when Cs(+) was used as charge carrier than when Na(+) and K(+) were used. In single-channel recordings with… Show more

“…C, Current-voltage relationships from experiments shown in A and B. The currents in the I-V plots are expressed as current densities which were calculated as described previously (Wesolowski et al, 2007) Among the four PMCA isoforms, PMCA1 and PMCA2 have been demonstrated to colocalize with PSD95 in the OPL Duncan et al, 2006). TMEM16B and PMCAs are absent from photoreceptor synapses of mice that lack MPP4, possibly because of the loss of PSD95 in the mutant terminals.…”

“…19,20 Gramicidin-hybrid channels were transferred into Chinese hamster ovary (CHO) cells and studied in vivo by patch clamp. 21 The Cs þ ion selectivity of compound 6 seen in BLM measurements could also be observed in CHO cells. For the case of tetrahydrofuranamino acids, the electrophysiological response of trabecular meshwork cells to tetrahydrofuran-gramicidin hybrid channels was investigated.…”

“…Tetrahydrofuran- and tetrahydropyran-amino acids could also be incorporated into the gramicidin motif to generate hybrid channels which were functionally characterized by conductance measurements. , Gramicidin-hybrid channels were transferred into Chinese hamster ovary (CHO) cells and studied in vivo by patch clamp . The Cs + ion selectivity of compound 6 seen in BLM measurements could also be observed in CHO cells.…”

“…Terminal-charged amphiphilic compounds, [7] can self-assemble into voltage-dependent pores and rigid push-pull rods [8] with permanent axial macrodipoles to give voltage-sensitive pores. Compared with these self-assembled pores, the channel-active b 6,3 -helix of gramicidin A (gA) is structurally well defined. [9] ( Figure 1)…”

“…In a continuation of our studies on the implantation of synthetic gramicidin-hybrid channels into cells (trabecular meshwork cells, [5] CHO cells [6] ), we turned to synthetic voltage-gated channels.
