Functional studies of the Synechocystis phycobilisomes organization by high performance liquid chromatography on line with a mass spectrometer

Abstract: This study was designed to yield data on the supramolecular organization of the phycobilisome apparatus from Synechocystis, and the possible effects of environmental stress on this arrangement. Phycobilisomes were dissociated in a low ionic strength solution and a quantitative estimation of the protein components present in each subcomplex was obtained using liquid chromatography coupled on-line with a mass spectrometer equipped with an electrospray ion source (ESI-MS). An advantage of this approach is that in… Show more

“…The molecular masses of a (16 kDa) and b (18 kDa) subunits of A. africanum APC (Fig. 2, lane 3) are lower than those of other cyanobacteria (Moreno et al 1997;Zolla et al 2002). Also, a few additional bands of some crude extract impurities other than C-PC were observed.…”

“…Recently we have described a non-chromatographic, rivanol-sulfate method for purification of C-PC from Spirulina (Arthrospira) fusiformis (Minkova et al 2003) as an attractive alternative to the procedures for purification of PBPs that usually combine fractional ammonium sulfate precipitation and various chromatographic methods (Moreno et al 1997;Zolla et al 2002;Patel et al 2005).…”

“…The chromophore in PC, phycocyanobilin, is attached by cysteinyl thioether linkages at positions 84 , 84 , and 155 in the and subunits. The supramolecular structures of PBSs have been determined for many organisms including Synechocystis, 13,14 Synechococcus, [15][16][17][18] and Anabaena. 11,19 However, the study of light transfer in PBSs remains a challenging task from both an experimental and theoretical point of view.…”

“…An approach for explaining the effect of UVB irradiation on fluorescence characteristics is given by Sinha et al (1995) and Zolla et al (2002). The pigment matrix of cyanobacteria is influenced by UVB-radiation.…”
