Functionalized Micellar Assemblies Prepared via Block Copolymers Synthesized by Living Free Radical Polymerization upon Peptide-Loaded Resins

Abstract: Hybrid peptidic-synthetic amphiphilic block copolymers, synthesized by living free radical polymerization (LFRP) on solid support, have been utilized as precursors for nanoscale materials possessing bio-available peptides. LFRP initiators, coupled to the peptide terminus upon the resin, facilitated the growth of homo- and block copolymers via nitroxide mediated radical polymerization (NMRP) or atom transfer radical polymerization (ATRP). Herein, the versatile solid-support synthesis of the antimicrobial peptid… Show more

“…[25][26][27][28][29][30] On the other hand, hydrophobic P(t-BA) can be easily functionalized by trans-esterification 31 or converted into biocompatible hydrophilic poly(acrylic acid). 32 The P(MOx) and P(t-BA) building blocks were synthesized by CROP and ATRP processes, respectively. The alkyne and azide terminal functionalizations were carefully characterized by MALDI-Tof spectroscopy and polymer-polymer coupling was then achieved using Huisgen's cycloaddition reaction.…”

“…and TP derivatized amphiphilic block copolymers [113]. Inhibition of a biofilm culture of Pseudomonas aeruginosa, or other oral pathogens, by the human host defense peptide LL-37 or a peptide mimictic based on the structure of MG 2 are examples of this application [114,115].…”

“…The viral HIV-I Tat protein transduction domain (PTD) sequence, YGRKKRRQRRR, was coupled onto the SCKs and was shown to then provide for cell transduction (21). Advanced synthetic methodologies were developed (22)(23)(24) and rigorous characterization studies were performed to determine the amount of PTD that was required to effect cell entry while maintaining biocompatibility (25). The role of nanoparticle shape was also explored, and it was found that small spherical particles entered cells to a greater extent than did elongated cylindrical particles (26).…”
