G-1639A but Not C1173TVKORC1Gene Polymorphism Is Related to Ischemic Stroke and Its Various Risk Factors in Ukrainian Population

Abstract: Vitamin K epoxide reductase complex subunit 1 (VKORC1) is integral 163-amino acid long transmembrane protein which mediates recycling of vitamin K 2,3-epoxide to vitamin K hydroquinone and it is necessary for activation of vitamin K-dependent proteins (VKDPs). Herein, the association between G-1639A (rs9923231) and C1173T (rs9934438) single-nucleotide polymorphisms (SNPs) of the VKORC1 gene and ischemic stroke (IS) was tested in Ukrainian population. Genotyping was performed in 170 IS patients and 124 control … Show more

“…Nonetheless, this limitation of our study is in direct relationship with its main strength, as our results stand as the first longitudinal study to assess the effect that VKORC1 and CYP4F2 polymorphisms have on boosting the risk of plaque development at 5 years. Until now, other studies have determined the role of this polymorphisms in a cross-sectional manner [12,30,35]. Therefore, the current study significantly advances the knowledge in the field of atherosclerotic plaque formation.…”

“…The presence of the rs9923231 polymorphism seriously impaired the function of VKORC1 and led to resistance to warfarin. 15,16 Patients with the rs9923231 G allele may have a different dosage versus effect relationship than wild-type patients. Therefore, these patients need a larger initiation dosage to affect anticoagulant time, and they may also need a larger dosage adjustment than the current routine dosage adjustment to bring an abnormal INR value into the target range.…”
