G-Trap Assay I: Small GTPases as sensitive immune response biomarkers for active bacteremia

Abstract: The annual toll of sepsis is a 33% mortality rate for hospitalized patients with a cost of greater than 60 billion dollars in the U.S. There is a correlation between sepsis mortality rates and time to treatment with broad-spectrum antibiotics. Consequently, antibiotics are prescribed to nearly every patient suspected of bacteremia. However, once determined that broad-spectrum antibiotics are not required, it is unclear how to optimize the de-escalation of the antibiotics. There is an urgent need for methods to… Show more

“…45, 46 Proinflammatory cytokines are frequently cited as bacteremia biomarkers in the literature. 2, 24, 47-52 In our previous study 1 we showed how this interplay between circulating pro-and anti-inflammatory cytokines impacted the magnitude of the correlated GTPase signals. Thus, GTPase activity in patient leukocytes represents an integrated signaling outcome reflecting the composition of circulating cytokines due to infection.…”

“…In a preceding study, 1 we used serial plasma samples from infected patients to simulate immune activation of human leukocytes and culture cells after exposure to inflammatory mediators released upon infection. That study showed that pro-/anti-inflammatory cytokines and novel biomarkers such as chemerin 24 induced GTPase activation in test cells.…”
