Gadd45α activity is the principal effector of Shigella mitochondria-dependent epithelial cell death in vitro and ex vivo

Abstract: Modulation of death is a pathogen strategy to establish residence and promote survival in host cells and tissues. Shigella spp. are human pathogens that invade colonic mucosa, where they provoke lesions caused by their ability to manipulate the host cell responses. Shigella spp. induce various types of cell death in different cell populations. However, they are equally able to protect host cells from death. Here, we have investigated on the molecular mechanisms and cell effectors governing the balance between … Show more

“…Indeed, bacteria do not proliferate within these cell populations, and they likely survive for a while. Shigella-infected epithelial cells also activate death programs (27,48,49). However, at the same time, these cells initiate a prosurvival response (27,50) allowing the bacteria to multiply and to colonize the microenvironment up until the death of the cells (51).…”

“…1F, S1B). S. flexneri infection is known to induce apoptosis in epithelial cells (Carneiro, Travassos et al, 2009, Lembo-Fazio, Nigro et al, 2011. However, the kinetic of activation of caspase-3 upon bacterial infection was not different between EGFP-RIPK2 expressing cells and unmodified HeLa cells ( Fig.…”

“…The knockdown of APE1 by siRNA increased the proliferation capacity of WI38 cells and also reduced the mRNA expression of senescence-related genes (e.g., p16, p21) ( Fig EV5F and G). Furthermore, colon samples from old people showed significantly higher percentage of GADD45A and c-H2AX-positive cells as compared to young ones ( Fig 6E-G and Appendix Table S1), and the upregulation of GADD45A is related to inflammation in human colon [39]. These data indicate that GADD45A and BER pathways participate in the replicative aging of human cells.…”
