Gain of function of stomatal movements in rooting Vitis vinifera L. plants: regulation by H2O2 is independent of ABA before the protruding of roots

Abstract: Reactive oxygen species (ROS), namely superoxide radical (O2(-)) and hydrogen peroxide (H(2)O(2)) are generated when plant tissues endure a variety of environmental stresses, including light stress. The extremely short life times of ROS makes the study of their production in planta very difficult. The use of ROS-specific tracer dyes, 3-3' diaminobenzidine and nitroblue tetrazolium, together with high-resolution imaging provides the opportunity to identify sites of photooxidative stress response by ROS accumula… Show more

“…This coincides with the protrusion of new roots and the expansion of the first ex vitro leaf and culminates with a peak of ABA and H 2 O 2 concentration on the seventh day after transfer, both produced in the newly-expanded and functional roots (Figure 3; Neves et al, 1998;Vilela et al, 2007). At this point, acclimatization to ex vitro is not yet complete but photooxidative stress is no longer a problem for the growing grapevine plants.…”

“…The heat map represents the differences between the values measured at the moment of transfer to ex vitro (control) and those monitored for 7 days of ex vitro growth: dark green, very significant increase from the control; light green, significant increase from the control; gray, no significant differences to the control; orange, significant decrease from the control; red, very significant decrease from the control. Values were retrieved from Carvalho et al (2006) and Vilela et al (2007).…”

“…The initial responses to ABA in stomatal closure in-clude increases in ROS (Vilela et al 2007), which are important signalling molecules in defense. This is in accordance with osmotic stress-induced strong inhibition of photosynthetic efficiency in grapevine leaf discs.…”
