Gambogic Acid Inhibits Angiogenesis and Prostate Tumor Growth by Suppressing Vascular Endothelial Growth Factor Receptor 2 Signaling

Abstract: Gambogic acid (GA), the main active compound of Gamboge hanburyi, has been previously reported to activate apoptosis in many types of cancer cell lines by targeting transferrin receptor and modulating nuclear factor-KB signaling pathway.

“…Conditioned medium from control adenovirus (dl312)-transduced MDA-MB-231 cells were able to induce tube formation of HCMECs within 18 h of incubation (Fig. 4A, a an b) as seen earlier (25,26). On the other hand, conditioned medium from AdMBP-1-transduced MDA-MB-231 cells significantly reduced tube formation (Fig.…”

“…The transwell migration assay was carried out as described previously (Yi et al, 2008). Briefly, HUVECs were seeded onto transwell first, and after the cells were attached, the medium in the bottom chambers was changed to Dulbecco's modified Eagle's medium with or without VEGF.…”

“…Xenograft mouse model was used to evaluate the effect of oridonin tumor growth as described by Yi et al (Yi et al, 2008). The 5-week-old to 6-week-old severe combined immune deficiency (SCID) male mice (SLAC Laboratory Animal, China) weighing about 20g were divided into four groups with 6 animals for each group: control group, 5-FU group, oridonin high-dose treated group and oridonin lowdose treated group.…”
