Gamma Interferon Does Not Enhance Clearance ofPseudomonas aeruginosabut Does Amplify a Proinflammatory Response in a Murine Model of Postseptic Immunosuppression

Abstract: Patients that have suffered a major injury may sustain a period of immunocompromise and altered Th1/Th2 cytokine balance that can predispose them to opportunistic infections. Pseudomonas aeruginosa is frequently a causative organism for nosocomial infections in critically ill patients and is associated with high mortality. We previously mimicked this clinical scenario by challenging mice with P. aeruginosa 5 days after a cecal ligation and puncture (CLP) procedure. Mice that were subjected to CLP had reduced a… Show more

“…The deleterious role of IFN-γ was demonstrated by its capacity to increase death when injected in mice, particularly when acting in synergy with TNF-α (12). This deleterious effect was also shown in the CLP model of polymicrobial peritonitis (92) and in a CLP model followed by a P. aeruginosa infection (93). Polymicrobial peritonitis performed in IFN-γ receptor-deficient mice (94) or in rats given anti-IFN-γ antibodies (95) led to the similar demonstration of this negative role.…”

“…Subjects with genetic deficiencies in IFN-c protein or receptor have increased susceptibility to mycobacterial infection [46,61]. Several families are reported to be vulnerable to disseminated MAC disease, and their immunocompromise is caused by the decrease in IFN-c production [62,63] or by IFN-c receptor abnormality [64].…”

“…Imbalances in this ratio may result in moderate to severe inhibition or functional alteration to critical immune system activities. It was previously demonstrated that trauma and sepsis favor a Th2-dominant status (17,18). As invasive surgeries often result in significant tissue trauma, the same conclusion may be drawn for surgical patients.…”
