Psychological stress produced by intraspecies emotional communication in a communication box was studied to see whether it could be applied to a new screening method for anti-ulcer agents. There were two groups of mice, the ‘sender’ mice that received electrical foot shocks and showed emotional responses such as piloerection, abnormal squealing and jumping, and the ‘responder’ mice that were affected by the sender’s emotional responses without foot shock. The gastric lesions (erosions) produced by conditioned emotional stimuli were observed in both groups. The effects of the anti-ulcer drugs cetraxate, cimetidine and gefarnate were examined. In ‘senders’, the gastric lesions were significantly suppressed by the administration of cetraxate at a dose of 200 mg/kg (p.o.) or cimetidine at doses of 30 and 100 mg/kg (p.o.). The gastric lesions in ‘responders’ were significantly suppressed by two administrations of cetraxate at doses of 100 and 200 mg/kg (p.o.), cimetidine at doses of 10–100 mg/kg (p.o.) or gefarnate at a dose of 200 mg/kg (p.o.). The gastric lesions of ‘responders’ were more sensitive to anti-ulcer drugs. The present results indicate that the gastric erosions of ‘responders’ are useful for the evaluation of anti-ulcer agents.

ABSTRACT-Gastric lesions were developed in the communication box paradigm (CB) in mice as well as in the activity-stress paradigm (AS) in rats. Treatment with sulpiride (10-320 mg/kg, p.o.) attenuated these psychological stress-induced gastric lesions in a dose-dependent manner, while it failed to suppress those induced by physical stress such as restraint water-immersion (WI) and indomethacin treatment (IND). In contrast, treatment with famotidine (0.32-10 mg/kg, p.o.) dose-dependently attenuated the gastric lesions induced by physical stress but not those by psychological stress. Pylorus-ligation study revealed that famotidine strongly reduced gastric acid secretion, whereas sulpiride minimally affected that. It was also demonstrated that physical stress (WI) enhanced acid secretion while psychological stress (CB and AS) rather depressed that. These results suggest that the mechanisms of gastric lesion formation are clearly different between physical and psychological stress and that sulpiride specifically attenuates psychological stress lesions possibly through a central mechanism.

“…Analises adicionais conduzidas com este modelo mostraram que o turnover de noradrenalina no SNC estava aumentado não apenas nos seanders, mas também nos responders confirmando a presença de estresse nestes animais(IIMORI et al, 1982). Neste sentido, fármacos ansiolíticos como os benzodiazepínicos mostraram-se capazes de reduzir a incidência de ulceração gástrica nos responders(ICHIMARU et al, 1984), assim como o do estímulo aversivo aos animais do grupo IS. Os autores observaram que os animais dos grupos IS e PS apresentaram aumento nos níveis de ansiedade medida no campo aberto e no pluz maze, diminuição da resposta imune inata e diminuição da resistência ao crescimento de um tumor ascítico de Ehrlich.…”
