Gastrodin Suppresses Pentylenetetrazole-Induced Seizures Progression by Modulating Oxidative Stress in Zebrafish

Abstract: Pentylenetetrazole (PTZ)-induced seizures in Zebrafish models are now widely accepted for investigating human disease epilepsy. In epilepsy, the generation of oxidative stress contributes to the brain injury. Although Gastrodin (GAS) has been reported to have anticonvulsant activities, its effects on zebrafish seizure models and the underlying mechanism remain unclear. In this study, we evaluated the effects of GAS pretreatment on PTZ-induced seizures in zebrafish larvae and investigated the underlying mechani… Show more

“…For PTZ treatment, PTZ (10mM) was added directly to the 96 well plate following acclimation period. Final concentration of PTZ was determined from previously published results (Baraban et al, 2005; Afrikanova et al, 2013; Peng et al, 2016; Jin et al, 2018).…”

“…The embryos were obtained from the natural mating of adult zebrafish bred then maintained in bathing water. Fertilized eggs were collected and transferred to 6-well plates (39). Plates were kept in an incubator with the temperature maintained at 28±0.5˚C and monitored daily until 2 days post-fertilization (dpf).…”
