GATA simple sequence repeats function as enhancer blocker boundaries

Abstract: Simple sequence repeats (SSRs) account for B3% of the human genome, but their functional significance still remains unclear. One of the prominent SSRs the GATA tetranucleotide repeat has preferentially accumulated in complex organisms. GATA repeats are particularly enriched on the human Y chromosome, and their non-random distribution and exclusive association with genes expressed during early development indicate their role in coordinated gene regulation. Here we show that GATA repeats have enhancer blocker ac… Show more

“…This (GATA) n motif is reported to be extended on Y or W chromosomes of several vertebrates, including human, mouse and snakes, and invertebrates species [3032]. Furthermore, (GATA) n motif (included in Bkm repeats with (GACA) n motifs) could be required for sex determination and differentiation as well as to participate in the higher order chromatin organization and function, particularly in the formation of heterochromatin [3335]. …”

“…The mtND1 200 bp PCR fragment was purified after PCR using 32 P dATP. For gel shift, radiolabeled DNA was incubated with mTSC extract for 30 min in 25 mm HEPES ( pH 7.6), 100 mm NaCl, 1 mm DTT, 0.1 mm PMSF, 10% glycerol, 10 μg tRNA and 0.5 μg poly(dI.dC) at room temperature following a previously described protocol (Kumar et al, 2013). For super-shift, equal amounts of TEAD4 antibody or IgG were included in the incubation mix.…”
