GATA3: a master of many trades in immune regulation

Abstract: GATA3 has conventionally been regarded as a transcription factor that drives the differentiation of T helper (Th) 2 cells. Increasing evidence points to function for GATA3 beyond controlling Th2 differentiation. GATA3 regulates T cell development, proliferation and maintenance. Furthermore, recent studies have demonstrated important roles for GATA3 in innate lymphoid cells. Thus GATA3 emerges as a factor with diverse functions in immune regulation, which are in some cases cell-type specific and in others share… Show more

“…We also observed frequent expression of GATA3 and BCL6 , though the relationship to function is not directly evident, as they have multiple effects on T cell function. GATA3 is associated with driving CD4 + Th2 responses, but is also involved in T cell homeostasis and Th9 differentiation [38]. BCL6 is associated with differentiation of follicular helper T cells, which have recently been identified as having a role in antibody-mediated cGVHD [39,40], and is also associated with multiple other effects, including CD8 + T cell proliferative burst and promoting memory T cell development [4143].…”

“…In the early sepsis stage, TCR trigger GATA-3 upregulation via PI3K-mTOR dependent pathways [41]. Moreover, GATA-3 is suggested to mediate notch signaling that is important for promoting T cell development and proliferation [42].…”

“…GATA3 is known to play crucial roles in a variety of immune cells in a developmental stage-and cell-lineage-specific fashion (27). For instance, GATA3 is important in the development and maintenance of T cells (27)(28)(29). Notably, GATA3 plays a critical role in the differentiation of Th2 cells and their ability to secrete cytokines such as IL-4, IL-5, and IL-13.…”
