Gaucher’s disease with Parkinson’s disease

Abstract: The association between type 1 Gaucher disease and PD has been reported in the literature. The clinical picture is characterized by the predominance of bilateral akinetic-rigid signs and poor response to levodopa therapy. The authors describe four patients (two siblings) with type 1 Gaucher disease presenting with the following signs of typical PD: asymmetric onset of rigidity, resting tremor, bradykinesia, and a favorable response to Parkinson therapies.

“…Sphingosine kinase is important for 1) revascularization responses, 2) regulating the maturation of vascular endothelial progenitors, and 3) controlling cellular recruitment (15,85,89). The cellular balance of the level of Cer, SPH, and S1P, termed the "sphingolipid rheostat," dictates cell fate, whereby Cer and SPH enhance apoptosis while the S1P promotes cell survival and proliferation (16,125,129). However, the actual sphingolipid rheostat may be broad, comprising numerous additional molecules in the SL metabolic map as well as those in related metabolic pathways.…”
