GCR1 Can Act Independently of Heterotrimeric G-Protein in Response to Brassinosteroids and Gibberellins in Arabidopsis Seed Germination

Abstract: Signal recognition by seven-transmembrane (7TM) cell-surface receptors is typically coupled by heterotrimeric G-proteins to downstream effectors in metazoan, fungal, and amoeboid cells. Some responses perceived by 7TM receptors in amoeboid cells and possibly in human cells can initiate downstream action independently of heterotrimeric G-proteins. Plants use heterotrimeric G-protein signaling in the regulation of growth and development, particularly in hormonal control of seed germination, but it is not yet cle… Show more

“…A few studies have also elucidated specific molecular mechanisms of hormonal cross-talk. They include the role of auxin and ethylene in regulating root meristem development (1), the antagonistic relationship between abscisic acid (ABA) and gibberellins (GAs) on seed dormancy and germination (5, 6), and integration of the primary signaling pathways of auxin and brassinosteroids (BRs) by auxin response factor 2 (ARF2) (7).Studies have indicated that BRs and ABA can coregulate the expression of hundreds of genes (4), and they interact physiologically in controlling many developmental processes (5,(8)(9)(10). It is also well known that ABA is required to establish seed dormancy during embryo maturation and to inhibit seed germination (5), whereas BRs promote seed germination, likely through enhancing the embryo growth potential to antagonize the effect of ABA (5, 10, 11).…”

“…Subsequent interactions between the receptors and their cognate proteins then drive signaling cascades resulting in biological outputs. Additionally, second messengers such as calcium, NO, ROS and G-proteins are intrinsically linked to most hormone responsive cellular processes (Lovegrove and Hooley 2000;Zentella et al 2002;Chen et al 2004;Higuchi et al 2004;Verslues and Zhu 2005;Chiwocha et al 2005;Okamoto et al 2006;Pandey et al 2006).…”

“…Sin embargo los estudios realizados en alelos de pérdida de función no implican a GCR1 de forma directa en la percepción de ABA. El mutante de pérdida de función gcr1 muestra hipersensibilidad a ABA, pero existen datos que confirman que GCR1 se encuentra en otras rutas de señalización por lo que tiene efectos pleiotrópicos [33].…”

“…Phosphatase assays were performed in a 50-mL reaction volume containing 20 mM Tris-HCl, pH 7.6, 10 mM MgCl 2 , 1 mM DTT, and ;10,000 cpm of 33 P-labeled casein. After incubation for 30 min at 308C, the reaction was stopped with 100 mL of 20% trichloroacetic acid, samples were centrifuged, and the release of 33 Pi in the supernatant was determined by scintillation counting.…”
