“…Techniques and Apparatus. A new gradient elution chromatography (86) has dual columns, a 9-chamber solvent programmer, and UV and differential refractometer detectors in series. A splitter inlet system (396), an automatic apparatus for loading a number of samples at preselected times (435), and other devices (119,236) for automating column chromatography have been described.…”

“…Nitrophenols have been separated on polyamidetreated thin-layers (143) or on silica gel G-amylopectin (430). Phenolic and heterocyclic fluorescent compounds have been studied by gel filtration chromatography on Sephadex G-15 (86).…”

“…22 Recently Evans et al 21 separated humic and fulvic acids in aquatic media and successfully introduced gel micro-samples by laser ablation for subsequent off-line ICP-MS determination of lead bound to humic and fulvic acid fractions. However, gel-based methods exhibit artifacts resulting from solute adsorption and chargerepulsion effects 24,25 and perhaps shearing of very large macromolecules 26 in the solid phase.…”

“…In control reactions (i.e., those without enzyme preparation, H 2 O 2 , or MnSO 4 ) the PCP was eluted as a broad peak around 150 ml, which was beyond the total effective column volume (Vt). Aromatic hydroxyl and methoxyl substituents are known to increase adsorption onto Sephadex (11). Therefore, elution of PCP in fractions beyond the Vt was probably due to interactions of the phenolic group with the Sephadex gel matrix.…”
