Gene expression profiles of bovine caruncular and intercaruncular endometrium at implantation

Abstract: At implantation the endometrium undergoes modifications necessary for its physical interactions with the trophoblast as well as the development of the conceptus. We aim to identify endometrial factors and pathways essential for a successful implantation in the caruncular (C) and the intercaruncular (IC) areas in cattle. Using a 13,257-element bovine oligonucleotide array, we established expression profiles at day 20 of the estrous cycle or pregnancy (implantation), revealing 446 and 1,295 differentially expres… Show more

“…Cluster analysis (Fig. 2) suggests that it might be possible to distinguish normal from abnormal conceptuses at d 18 (i.e., two outlier AI embryos compared with clustering of most of the SCNT clones), which could lead to improved methods for SCNT by varying conditions during the deprogramming and reprogramming phases (26,36).…”

“…The IFNT protein has antiluteolytic activity (2) and has been shown to have dramatic effects on gene expression in bovine primary endometrial cell cultures (36). The observed down-regulation of genes involved in IFNT signaling at d 18 in EET may disrupt paracrine pathways between EETs and the endometrium and potentially have direct effects on the adhesion of the trophoblast to the endometrium (SI Appendix, Fig.…”

“…In this study, the protein levels of PTN increased with the increase of NTS in EECs. Considering the fact that PTN mainly expressed in the caruncular areas of bovine endometrium [58], it might participate in the proliferation of endometrial cells in dairy goats.…”

“…Together with progesterone (P4), IFNT regulates endometrial gene expression necessary for the establishment of the proper uterine environment during the implantation period (Klein et al, 2006). A panel of approximately 30 genes was identified as expressed on day 16 as part of the early endometrial response to the conceptus and may represent early endometrial markers of a viable preimplantation conceptus , Mansouri-Attia et al, 2009a, reviewed by Lonergan and Forde . Although most data demonstrates that the main molecule affecting the endometrium is IFNT, additional conceptus secreted molecules, including prostaglandins (PG; Dorniak et al, 2012, Spencer et al, 2013 and cortisol (Dorniak et al, 2013b), also act on the endometrium.…”
