Gene Expression Profiling in Laser-Microdissected Bone Marrow Megakaryocytes

Abstract: Gene expression analysis of the megakaryocytic lineage requires isolation of megakaryocytes from their bone marrow microenvironment. Laser microdissection of megakaryocytes from diagnostic bone marrow biopsies allows analysis of standardised formalin-fixed samples that reflect the in situ grown status quo of a physiological or pathological condition. Taking into account that in neoplastic proliferation, e.g. myeloproliferative neoplasms, non-neoplastic haematopoietic clones proliferate in parallel, this morpho… Show more

“…Laser microdissection of single cells was performed as described by Hussein (20). In brief, 6 µm cryosections of the reconstituted liver were transferred onto lysine-coated ultraviolet-light-treated membrane slides (MMI) and put on a fluorescence microscope with a SmartCut Plus laser microdissection device (MMI).…”

“…71 These, in turn, led to the proposal of molecular classifications for AML 72 and myeloproliferative neoplasms, 73 and, more importantly, to multi-gene panels for mutation assessment in clinical practice. 74 In recent years, further technical improvements (e.g., single-cell analysis, molecular characterisation of laser micro-dissected samples, and gene expression profiling based on RNAsequencing [75][76][77] ) have literally revolutionised BM studies, questioning the value of traditional morphology and phenotypic assessment. 78 Nevertheless, there are many reasons why BM examination is still relevant to our daily clinical practice.…”

“…Laser capture micro-dissection (LCM) enables the isolation of single cells or small cell groups from histological sections under direct microscopic control [ 1 ]. Combined with quantitative PCR, LCM is a very powerful approach for studying individual transcript expression profiles in discrete cell populations [ 2 ]. Especially for renal biopsies, micro-dissection of its different compartments—glomeruli, tubulointerstitium and arteries/arterioles—is a worthwhile exercise despite its small amounts of degraded RNA.…”
