Generation and characterization of new CCR5-tropic HIV-1rmt clones

Abstract: : To develop effective non -human primate models for coping with numerous HIV-1/AIDS studies, rhesus macaque-tropic HIV-1 (HIV-1rmt) clones with a variety of biological properties are required. Such clones, if available, are powerful tools to experimentally elucidate HIV-1 replication and pathogenicity in host individuals, and also to develop anti-HIV-1 drugs/vaccines. However, only limited numbers of HIV-1rmt clones have been currently reported. In the present study, we generated new HIV-1rmt clones carrying … Show more

“…However, MN5/LSDQgtu replicated poorly compared to MN4/LSDQgtu in RM cell line (Doi et al, 2013). Additional CCR5-tropic clones carrying pSHIV AD8−EH or env clones from clinical isolates have been generated in the backbone of MN5/LSDQgtu (Doi et al, 2017). Interestingly, two of the clones carrying env genes from clinical isolates, designated gtu + Cl1 and gtu + A4Cl1, grew better than the parental clone MN5/LSDQgtu (or 5gtu) in RM M1.3S cells.…”

“…Interestingly, two of the clones carrying env genes from clinical isolates, designated gtu + Cl1 and gtu + A4Cl1, grew better than the parental clone MN5/LSDQgtu (or 5gtu) in RM M1.3S cells. Furthermore, gtu+A4Cl1 grew comparably well with MN4/LSDQgtu in PBMCs isolated from two different donor RMs (Doi et al, 2017). To determine the replication potential in vivo, two RMs were challenged with CXCR4-tropic MN4/LSDQgtu and one RM with CCR5-tropic gtu+A4Cl1 (Doi et al, 2018).…”

“…From the prototype HIV-1mt designated NL-DT5R ( Kamada et al, 2006 ), we have modified its genome stepwise and improved its replication potential in macaque cells through sequentially introducing necessary mutations/variations by in vitro mutagenesis coupled with viral genome adaptation in cells ( Nomaguchi et al, 2008 , 2011 , 2013a , b , c ; Kuroishi et al, 2009 ; Saito et al, 2011 ; Doi et al, 2017 ). Of particular note and importance, we resultantly have obtained CXCR4-tropic and CCR5-tropic HIV-1rmt (rhesus macaque-tropic) clones that grow well in rhesus cells ( Nomaguchi et al, 2013b , 2014 ; Doi et al, 2017 ). Figure 1 shows the basic genome structure of our HIV-1rmt clones.…”

“…Genomic regions of HIV-1rmt clones derived from HIV-1 NL4-3, SIVmac MA239, and SIVgsn 166 (SIV isolated from the greater spot-nosed monkey) are depicted by white, blue, and orange areas, respectively. Generation and characterization of HIV-1 NL4-3 ( Adachi et al, 1986 ), SIVmac MA239 ( Shibata et al, 1991 ), CXCR4-tropic HIV-1rmt designated MN4/LSDQgtu ( Nomaguchi et al, 2013b ; Nomaguchi et al, 2017 ), CCR5-tropic HIV-1rmt designated MN5/LSDQgtu ( Nomaguchi et al, 2013b ; Nomaguchi et al, 2017 ), and CCR5-tropic HIV-1rmt designated gtu + A4CI1 ( Doi et al, 2017 ) have been fully described previously. MN4/LSDQgtu and MN5/LSDQgtu carry growth-enhancing mutations in Gag-capsid, Pol-integrase, and Env regions as previously described ( Nomaguchi et al, 2013a , b ).…”
