Generation of cell-mediated cytotoxicity against bovine-papilloma-virus-transformed primary mouse cell lines

Abstract: The immunogenicity and immunosensitivity of primary mouse cell lines transformed by bovine papilloma virus 1 (BPV1) DNA were studied in a syngeneic mouse model by determining cell-mediated cytotoxicity in the spleens of mice immunized with the transformed cells. One of the cell lines induced the generation of cell-line-specific Thy1.2-positive cytotoxic effector cells. However, most of the cell lines tested induced the generation of Thy1.2-positive effector cells, which in addition to BPV1-transformed cells we… Show more

“…Without IFNy treatment of the target cells, this cytotoxicity is only detectable if the spleen cells from B6 mice immunized with BPVl-transformed cell lines are secondarily stimulated in vitro with irradiated BPVl-transformed cells [6]. However, if the target cells were treated with IFNy, cytotoxic activity was also detected in control cultures in which the immune spleen cells were incubated in the absence of BPVl-transformed stimulator cells.…”

“…In previous studies we have shown that the B6B31.CNuT.A cell line, in addition to inducing the generation of LAK-type effector cells, also induces the generation of cell-line-specific cytotoxic T lymphocytes [6]. In the present study, B6B31.C-NuT.A cells were used to study the effect of 1FNy on the immunogenicity of BPVl-transformed cells.…”

“…Discussion also showed exceptional reactivity in an in vitro cytotoxicity assay [12]. The low tumorigenicity of B6B22-NuT.A cells was in agreement with the history of the cell line; it was derived from a late-appearing tumor with a low degree of malignancy as determined by histology [ 15].…”
