Generation of Gene-Specific Mutated Rats Using Zinc-Finger Nucleases

Abstract: The genetic dissection of physiological and pathological traits in laboratory model organisms is accelerated by the ability to engineer loss-of-function mutations at investigator-specified loci. This chapter describes the use of zinc-finger nucleases (ZFNs) for the targeted disruption of endogenous rat genes directly in the embryo. ZFNs can specifically disrupt target genes in cultured rat cells and in embryos from inbred and outbred strains, leading to permanently genetically modified animals. This technology… Show more

“…To directly assess the importance of p67 phox in the saltinduced hypertension of the SS rat, site-directed mutagenesis by the application of zinc-finger nuclease technology (58,59) was used to create SS rats containing a null mutation of the p67 phox gene (51). As shown in Fig.…”

“…Mutant SS rat strains were generated by microinjection of custom CompoZr ZFNs (Sigma) into one-cell SS/JrHsdMcwi (SS) rat embryos and then implanted into pseudopregnant Sprague Dawley females, as described previously (Geurts et al 2010). Supplemental Table 2 provides the strain designations, target sites, DNA recognition helices of ZFNs, and description of gene mutation.…”

“…Recently, zinc finger nuclease technology (ZFN) was described as an efficient means to generate targeted mutations in rats (8,9,19). To eliminate the potential side effects of pharmacological agents and to address the role of the immune system in hypertension and renal disease in a genetic model of disease, a ZFN was designed to target recombination-activating gene 1 (Rag1) in the Dahl SS genetic background.…”
