Generation of Leukotriene B4by Human Lung Fragments and Purified Human Lung Mast Cells

Abstract: Leukotriene B4 (LTB4) is a potent stimulus for neutrophil chemotaxis, aggregation, and activation. Although this mediator has been postulated as a possible stimulus for the neutrophil accumulation seen after mast cell triggering in vivo, the ability of human mast cells to produce this leukotriene has never been described. The purpose of this study was to investigate the ability of purified human lung mast cells and mast cells in lung fragments to generate leukotriene B4 after immunologic (anti-IgE) and nonimmu… Show more

“…Calcium ionophore and concanavalin A in duced a time-and dose-dependent histamine release. In agreement with previous reports [18,29,39,40], histamine release induced by the calcium ionophore was more pro nounced than that by concanavalin A reaching maximally 46.4 and 15% of total histamine content, respectively. Compared to isolated human lung mast cells, histamine release in the lung fragments was slower.…”

“…Nasal antigenic provocation has demonstrated the in vivo formation of LTC4/D4/E4 at levels of 8,270 ±2,340 pg/ml sample [21]. Lcukotriene B4 is detect ed in relatively low concentrations following anti gen challenge of both lung mast cells and the nasal mucosa [34]. This supports the hypothesis that mast cells are a contributory source of leukotrienes during the early allergic response, but other cells arc also known to generate LTC4.…”

“…These cells release several mediators which have neutrophil chemotactic activity, including LTB4 and che motactic cytokines [13,14]. Recently, we demonstrated that IL-8, a potent leukocyte chemoattractant, induces in vivo neutrophil migration by stimulating the release of a neutro phil chemotactic factor from mast cells [15].…”
