Researchers from a broad spectrum of scientific and engineering disciplines are increasingly using scattering‐type near‐field infrared spectroscopic techniques to characterize materials non‐destructively with nanoscale spatial resolution. However, a sub‐optimal understanding of a technique's implementation can complicate data interpretation and act as a barrier to entering the field. Here the key detection and processing steps involved in producing scattering‐type near‐field nanoscale Fourier transform infrared spectra (nano‐FTIR) are outlined. The self‐contained mathematical and experimental work derives and explains: i) how normalized complex‐valued nano‐FTIR spectra are generated, ii) why the real and imaginary components of spectra qualitatively relate to dispersion and absorption respectively, iii) a new and generally valid equation for spectra which can be used as a springboard for additional modeling of the scattering processes, and iv) an algebraic expression that can be used to extract an approximation to the sample's local extinction coefficient from nano‐FTIR. The algebraic model for weak oscillators is validated with nano‐FTIR and attenuated total reflectance Fourier transform infrared (ATR‐FTIR) spectra on samples of polystyrene and Kapton and further provides a pedagogical pathway to cementing some of the technique's key qualitative attributes.

Mesoporous silica nanoparticles (MSNPs) are promising nanomedicine vehicles due to their biocompatibility and ability to carry large cargoes. It is critical in nanomedicine development to be able to map their uptake in cells, including distinguishing surface associated MSNPs from those that are embedded or internalized into cells. Conventional nanoscale imaging techniques, such as electron and fluorescence microscopies, however, generally require the use of stains and labels to image both the biological material and the nanomedicines, which can interfere with the biological processes at play. We demonstrate an alternative imaging technique for investigating the interactions between cells and nanostructures, scatteringtype scanning near-field optical microscopy (s-SNOM). s-SNOM combines the chemical sensitivity of infrared spectroscopy with the nanoscale spatial resolving power of scanning probe microscopy. We use the technique to chemically map the uptake of MSNPs in whole human glioblastoma cells and show that the simultaneously acquired topographical information can provide the embedding status of the MSNPs. We focus our imaging efforts on the lamellipodia and filopodia structures at the peripheries of the cells due to their significance in cancer invasiveness.

THz scattering-type scanning near-field optical microscopy (THz s-SNOM) enables high-resolution nanoscale 2D imaging, crucial for various disciplines including biology, physics, and materials science. This study establishes a reliable 3D model to determine the maximum thickness detectable by the probe. The influences of the demodulation order, tip radius, tip vibration amplitude, and incident frequency on the maximum detectable thickness are analyzed. Using bilayer samples as examples, we successfully inverted the thickness of the first layer within the detectable maximum depth range. However, we found that inversion fails when the thickness of the first layer is too small. This underscores the distinct difference between vertical and lateral resolutions, where vertical resolution represents the minimum calculable thickness. This research unveils complex internal structures, laying the groundwork for future nanolayer imaging.
