Genetic and immune characteristics of sentinel lymph node metastases and multiple lymph node metastases compared to their matched primary breast tumours

Abstract: Background Patients with breast cancer presenting with single lymph node metastasis (from a sentinel node) experience prolonged survival compared to patients with multiple lymph node metastases (≥3). However, little information is available on the genetic and immunological characteristics of breast cancer metastases within the regional lymph nodes as they progress from the sentinel lymph node (SLN) downstream to multiple regional lymph nodes (MLNs). Methods Genomic prof… Show more

“…In our study, the median TMB was 5.67 muts/mb in primary tumor group and 3.91 muts/mb in lymph nodes group. Lymph node TMB was generally lower than that of the primary tumor, suggesting that the efficacy of ESCC lymph nodes receiving immunotherapy may be inferior to that of the primary lesion, which is consistent with the reports of breast cancer and lung cancer 32,33 …”

“…Lymph node TMB was generally lower than that of the primary tumor, suggesting that the efficacy of ESCC lymph nodes receiving immunotherapy may be inferior to that of the primary lesion, which is consistent with the reports of breast cancer and lung cancer. 32,33 A growing body of research indicates that the tumor microenvironment is essential for immune response regulation, immune escape promotion, angiogenesis facilitation, and metastasis induction. 34,35 Numerous measures have been found to describe the ability of tumor cells to trigger adaptive immunological responses, the immune activation's downstream effects, like the existence of immune cells, and the immune scores' gene expression profile.…”

“…As the major component of TIICs, TAMs were well-known to promote distant metastasis by multiple mechanisms ( 39 , 40 ), and current studies found that the tumor cells present in lymph nodes maybe an important source of systemic metastasis ( 41 , 42 ). In the most recent study by Chen et al, TAMs were found to significantly higher in breast primary tumor with lymph node metastasis compared with tumor tissues without lymph node involvement ( 43 ).…”

“…EMT was one of malignant characteristic and a key foundation of cancer progression. 12 , 13 , 40 Twist1 signaling boosted tumor progression and promoted metastasis in ccRCC. 41 G2M checkpoint had a significant effect on cell proliferation, especially tumors.…”

“…11 Epithelialmesenchymal transition (EMT), is a characteristic of malignant tumor. [12][13][14] Inhibition of tumor EMT can slow down tumor metastasis. 15,16 Target of tyrosine kinase inhibitor (TKI) drugs had been discovered for treatment of metastatic ccRCC patients, 17,18 but it still leads to poor progression and recurrence when incomplete reversal of tumor hypoxia and EMT in ccRCC.…”
