ABSTRACT. Previous studies have shown that cytokines can affect serum lipoprotein concentrations. The aim of this study was to examine the association between IL-10 gene polymorphisms and serum lipoprotein levels of Han Chinese individuals. A total of 359 Han Chinese people were enrolled in this investigation. IL-10 -592, -819, and -1082 genotypes were established using polymerase chain reaction-restriction fragment length polymorphism analysis. An automatic biochemistry analyzer was used to determine serum concentrations of total cholesterol (TC), triglycerides (TG), low-density lipoprotein (LDL), high-density lipoprotein (HDL), and very low-density lipoprotein (VLDL) in each individual. We observed that the three IL-10 polymorphisms did not significantly differ in terms of age or age of carrier (P > 0.05), and the -592 and -819 variants did not significantly affect serum lipoprotein levels (P > 0.05). HDL concentrations were higher and TG levels were lower in carriers of the -1082 GA genotype compared to those with the AA genotype, and these differences were statistically significant (P < 0.05). Our results suggest that the polymorphism at position -1082 in the promoter region of IL-10 may affect serum HDL and TG concentrations, while other variants of this gene appear to have no relationship with serum lipoprotein levels.

Primary biliary cirrhosis (PBC) is a chronic autoimmune cholestatic liver disease most commonly encountered in postmenopausal women; it is characterized by high-titer serum autoantibodies to mitochondrial antigens, elevated serum IgM, progressive destruction of intrahepatic bile ducts, and ultimately liver cirrhosis and failure. The cytopathic mechanisms leading to the selective destruction of intrahepatic cholangiocytes are still largely unknown. The current theory on the pathogenesis of PBC indicated that environmental factors might trigger autoimmunity in genetically susceptible individuals. In fact, genetic predisposition is critical to disease onset and progression, yet peculiar among autoimmune diseases, as indicated by the lack of a strong association with major histocompatibility complex haplotypes. Further, the recently reported concordance rate among monozygotic twins strenghtens the importance of genetic factors, while also indicating that additional factors, possibly infectious agents or xenobiotics, intervene to trigger the disease. In this review, the available data regarding the genetic factors associated with PBC susceptibility and progression, as well as the available evidence regarding the immunomediated pathogenesis of PBC, will be critically illustrated and discussed.

The concept of genetic susceptibility in the contribution to human disease is not new. What is new is the emerging ability of the field of genomics to detect, assess, and interpret genetic variation in the study of susceptibility to development of disease. Deciphering the human genome sequence and the publication of the human haplotype map are key elements of this effort. However, we are only beginning to understand the contribution of genetic predisposition to complex liver disease through its interaction with environmental risk factors. In the coming decade, we anticipate the development of human studies to better dissect the genotype/phenotype relationship of complex liver diseases. This endeavor will require large, well-phenotyped patient populations of each disease of interest and proper study designs aimed at answering important questions of hepatic disease prognosis, pathogenesis, and treatment. Teamwork between patients, physicians, and genomics scientists can ensure that this opportunity leads to important biological discoveries and improved treatment of complex disease.
