Genetic characterization of enterovirus 71 isolated from patients with severe disease by comparative analysis of complete genomes

Abstract: Enterovirus 71 (EV71) which causes mild illness in children is also associated with severe neurological complications. This study analyzed the complete genomes of EV71 strains derived from mild and severe diseases in order to determine whether the differences of EV71 genomes were responsible for different clinical presentations. Compared to complete genomes of EV71 strains derived from mild cases (less virulent strains), nucleotide differences in EV71 strains isolated from severe cases (more virulent strains) … Show more

“…The mature capsid is built of 60 structural subunits or protomers, which are comprised of four structural proteins, VP1 to VP4 (VP1-4). Several key residues that map to the 5-fold symmetry vertex, VP1 98 and 145, have been linked to positive selection, virulence, and receptor binding (4,(10)(11)(12).…”

“…6) (16, 22). VP1-145 has been found to be under positive selection (36,37) and has been implicated as one of the possible determinants of virulence in humans (38,39) and adaptation in the mouse (40)(41)(42). It is likely that the EV71 antigenicity, as well as receptor-binding phenotype, is altered with a single mutation at VP1-145, which may confer an advantage to EV71 for effective host infection and transmission.…”

“…A nonconservative amino acid change in VP1 located within the BC loop (L97R) contributes to dissemination and neurotropism in immunocompromised patients (24). The residue at position 145 of VP1 (VP1-145) controls virus tropism by changing the accessibility of the positively charged lysine side chain of VP1-244 to the negatively charged N terminus of PSGL-1 on leukocytes (25) and has been implicated as one of the possible determinants of virulence in humans (26,27). Moreover, VP1 is an attractive target for identification of EV71 inhibitors.…”
