Genetic diversity analysis of some Egyptian Origanum and Thymus species using AFLP markers

Abstract: BackgroundIn the present investigation, two genera of family Labiatae (genus Origanum and genus Thymus including their available species in Egypt) were analyzed genetically on DNA level using amplified fragment length polymorphism (AFLP) markers. Four selective primer combinations (E-AGC/M-CAA, E-AGC/M-CAC, E-AGG/M-CTC, and E-ACC/M-CAT) were used to detect genetic variations (polymorphisms) within and between the studied plant species of each genus and with their cultivated relatives.ResultsA total of 193 ampl… Show more

“…The values of polymorphic information content represent the level of marker effectivity in the determination of polymorphism in the set of genotypes. The value of PIC depends not only on the number of produced fragments but also on their relative frequency [50,51]. On average, we detected higher average values of PIC in the SCoT markers (0.795) in comparison to the RAPD markers (0.760).…”

“…Molecular markers have become increasingly important in evaluating species diversity and evolutionary relationships [15]. For researchers, the genetic analysis of plants is a foundation for characterizing natural plant genetic resource, detecting genetic diversity or genetic homogeneity, and selecting plants with specific traits such as the synthesis of desired chemicals and stress tolerance mechanisms [8]. Salicornia consists of approximately 15 genus and 68 species [29].…”

“…It involves preselective amplification followed by selective amplification that generates specific molecular markers for a particular sample. AFLP analysis is a robust, multi-locus PCR-based DNA fingerprinting technique that can provide the most efficient, reliable, and economical population genetics analysis (El-Demerdash et al, 2019). AFLPs detect nuclear DNA markers inherited in a Mendelian fashion and provide a much greater level of polymorphism that cover a wider genomic area when compared to RAPD and ISSR (Zhao et al, 2018).…”
