Genetic diversity and fingerprinting of 33 standard flue-cured tobacco varieties for use in distinctness, uniformity, and stability testing

Abstract: Background: At present, the distinctness, uniformity, and stability (DUS) testing of flue-cured tobacco (Nicotiana tabacum L.) depends on field morphological identification, which is problematic in that it is labor intensive, timeconsuming, and susceptible to environmental impacts. In order to improve the efficiency and accuracy of tobacco DUS testing, the development of a molecular marker-based method for genetic diversity identification is urgently needed. Results: In total, 91 simple sequence repeats (SSR) … Show more

“…The PIC value for InDel markers developed by RAD-seq in tobacco was higher than 0.33 in sesame (Kizil et al 2020) but lower than 0.56 in rice (Liu et al 2015). Our PIC values are reasonable for evaluating the genetic relationship among tobacco accessions, according to He et al (2020). Analysis of the genetic diversity also revealed diversity and I values of 0.48 and 0.34, respectively (Table S3), which are similar to previous studies (Fan et al 2016;He et al 2020).…”

“…The 47-SNP barcode is rich in variations and has high power to discriminate the 216 cigar tobacco accessions from one another. On average, one SNP locus can identify five accessions, a resolution capacity that is much higher than that reported for ISSRs (Yang et al, 2010 ) and SSRs (He et al, 2020 ), which have been used for the identification and evaluation of tobacco accessions in the past. A SNP barcode provides a tool to discriminate very closely related accessions or the origins of the cigar tobacco subgenomes.…”

“…DNA fingerprinting has the advantages of being fast and accurate. In addition, DNA fingerprinting is a powerful tool for identifying varieties and strains, and it is also well suited for the identification of plant germplasm resources (Sorkheh et al, 2007 ; Gramazio et al, 2017 ; He et al, 2020 ; Wu et al, 2020 ). SNP marker-based high-throughput detection technology and data analysis is mainly based on the genetic characteristics of its “binary variation”—using two different fluorophores to label two different allelic variants—and a fluorescence detection system that can effectively distinguish two homozygous and one heterozygous genotypes.…”

“…They have been recognized as one of the most powerful marker systems for identifying plant variety and have been successfully applied across multiple species [8,44]. For instance, He et al [45] established the genetic ngerprints of 33 standard ue-cured tobacco varieties using 48 SSR markers and developed identi cation technology for new tobacco varieties based on SSR markers. Chen et al [44] created a DNA ngerprinting database of 128 excellent oil camellia varieties using highly variable SSR markers.…”
