Genetic Diversity and Phylogeny of Rhizobia That Nodulate Acacia spp. in Morocco Assessed by Analysis of rRNA Genes

Abstract: Forty rhizobia nodulating four Acacia species (A. gummifera, A. raddiana, A. cyanophylla, and A. horrida) were isolated from different sites in Morocco. These rhizobia were compared by analyzing both the 16S rRNA gene (rDNA) and the 16S-23S rRNA spacer by PCR with restriction fragment length polymorphism (RFLP) analysis. Analysis of the length of 16S-23S spacer showed a considerable diversity within these microsymbionts, but RFLP analysis of the amplified spacer revealed no additional heterogeneity. Three clus… Show more

“…The fact that bacteria isolated from the nodules of legume plants is similar to Agrobacteria has already been reported (Tan et al 1999). Khbaya et al (1998), and also explains why some of the strains present in the nodules that resembled Agrobacterium could not nodulate the original host. However, bacteria isolated from Caragana intermedia could induce nodule formation in hosts after infected Caragana intermedia for one month.…”

“…This strain could be useful for the inoculation and improvement of A. seyal-rhizobium symbiosis in salt-affected soils (Diouf et al 2005). African acacias are predominantly nodulated by Mesorhizobium, Rhizobium, or Ensifer genera (Haukka et al 1998;Khbaya et al 1998;de Lajudie et al 1998;Ba et al 2002;Sarr et al 2005;Diouf et al 2007 Figure 4 Phylogenetic (neighbour-joining) tree based on 362 nt aligned sequences of nifH gene using MEGA4. “…The length of the ampli®ed region, which included almost the entire 16S RNA gene (1500 bp; Willems and Collins 1993), the IGS and about 100 bp of the 23S rDNA, was different among the S. junceum isolates and also among the reference strains ( Fig. 1 and Table 1) as expected (Laguerre et al 1996;Khbaya et al 1998).…”

“…The combination of the 16S rRNA gene and 16S±23S IGS generated a long and variable (Fig. 1) PCR fragment whose size increased the potential to differentiate between close strains (Nour et al 1994a;Gu Èrtler and Stanisich 1996;Laguerre et al 1996;Normand et al 1996;Selenska-Pobell et al 1996;Khbaya et al 1998;de Oliveira et al 1999).…”
