Physical barriers like head works, dams barrages are main cause of fragmentation and declining of freshwater fish population in natural habitat. Present study focused on RAPD marker technique to assess the genetic variability among and between the populations of endangered sperata sarwari (Singharee) inhabited in the Indus drainage system of Punjab. Total eight populations (80 speciemen) of S. sarwari were collected from the up and downstream of four Rivers (Chenab, Jhelum, Ravi and Indus) of Punjab. Genomic DNA isolated from muscle tissue of adipose side and ten RAPD marker were used, which produced 50 scorable bands with average band ranging from 250–1050 bp which used for further genetic analysis of S. sarwari. Downstream Indus population of S. sarwari showed highest values of observed alleles (na), effected alleles (ne), Nei’s diversity (h), Shannon index (I) and polymorphism which indicated that downstream Indus population was more genetically variant. The genetic variability (0.5124) and genetic flow (0.4758) among the eight population S. sawari was observed. The up and downstream population showed the highest genetic distance (0.5738) and lowest genetic similarities (0.5634) which indicated complete isolation of Ravi population from other six population. Dendrogram showed that up and downstream Ravi population was completely isolated from the other six up and down stream population of S. sarwai. Overall results indicated that the presence of high fragmentation in River Ravi caused the destruction of habitat and decline in population of S. sarwai in the River Ravi.

The freshwater air-breathing swamp eel Monopterus spp. are native to the freshwater of Bangladesh and throughout the Indian subcontinent. To identify the different swamp eel species and to check the genetic diversity among them, a total of twelve swamp eel specimens were collected from four districts (Tangail, Bogura, Bagerhat and Sylhet) representing the four division of Bangladesh. The extracted DNA from twelve fish samples was amplified by the PCR technique for DNA barcoding and RAPD analysis. Among 12 specimens, 8 specimens showed a 95-100% similarity with M. cuchia species published in the NCBI GenBank database and BOLD system. The studied mct3 (collected from Tangail region), mcs1, mcs2 and mcs3 (collected from Sylhet region) specimens showed about 83% homology with Ophisternon sp. MFIV306-10 as per BLAST search; whereas BOLD private database showed 99% similarity with Ophisternon bengalense (Bengal eel). From the phylogenetic tree analysis, 8 samples were clustered with M. cuchia and 4 samples showed similarity with Ophisternon sp. MFIV306-10 and Ophisternon bengalense _ANGBF45828-19. In RAPD-PCR based analysis, it was found that the maximum genetic distance (1.6094) was observed between mcba2 and mcs3, while between mct1 and mct2, the minimum genetic distance was 0.000. A total of 192 bands, of which 35 were polymorphic with 17.88% polymorphisms among swamp eel species and the size of the amplified DNA fragments ranged from 250 to 1800 bp. The information on DNA barcoding and RAPD analysis help measure the genetic diversity among swamp eel species, ensure the reliability of the published taxonomic information, and initiate proper management programs to conserve these vulnerable species to meet future export demand. The wild stock of Indian major carps is declining in major rivers of India due to pollution, overexploitation, and inbreeding depression. In the present study, the comparative population structure, genetic diversity, and historical demographics of IMCs, Labeo rohita, Labeo catla, and Cirrhinus mrigala were characterized by analyzing partial 307bp sequences of Cytochrome b gene of 357 individuals collected from seven geographically isolated sites from Indian river basins and two culture zones. The DNA sequences alignment among three species revealed comparative divergence and evolution in these heterogeneous species in terms of over transition to transversion ratio of 6.25. The study implicated the occurrence of 35 haplotypes with haplotype diversity of 0.7333 in L. rohita at Brahmaputra basin showing allelic richness, while no nucleotide diversity (π) 0.000 was observed in L. catla of two geographically isolated locations of Rivers Ganga and Narmada. The analysis of molecular variance (AMOVA) revealed the genetic diversity of IMCs is to be very low (9.40%) within the species, as compared to among the three species (80.76%). The Fst scores ranged between 0.000 and 0.996, indicating low to high genetic structure of IMCs. The Haplotype-1, Haplotype-13, and Haplotype-19 in L. rohita, L. catla, and C. mrigala were considered as ancestral haplotypes, respectively. The expansion factor was found to be 0.45000 and 0.90000 for L. rohita stocks at Farakka-Ganga and Allahabad-Ganga, respectively. The present study provides the first report on genealogical biogeography, evolutionary divergence, and expansion of three IMCs, which could help to formulate the strategies for conservation of fisheries biodiversity in the Indian riverine ecosystems.
