Although hydroelectric power plants generate most electricity in Brazil, the dams provide environmental barriers which also include the interruption of fish migration. Current study, conducted prior to the construction of São Domingos HPP on the shaft of the Cachoeira Branca (waterfall) on the Verde river, state of Mato Grosso do Sul, Brazil, evaluated the genetic diversity of two natural populations of dourado (Salminus brasiliensis) located downstream (Population A -PopA) and upstream (Population B -PopB) the waterfall. Eight primers were employed to analyze 56 specimens (PopA: 26; PopB: 30) and observed in 102 fragments, of which 86 were polymorphic (84.3%). Low frequency fragments (PopA: 2; PopB: 1), three limiting fragments (PopA) and three exclusive fragments (PopB) were identified. The intra-population genetic variability calculated the Shannon index, whilst the percentage of polymorphic fragments showed high rate variability within each population (PopA: 0.300 and 60.80% and PopB: 0.411 and 79.40%, respectively). The distance and genetic identity revealed high genetic differentiation (0.076 and 0.927, respectively). Results reveal that populations have high intra-population genetic variability and genetic differentiation between them, with low gene flow. ResumoAs usinas hidrelétricas são responsáveis por gerar a maioria da energia elétrica utilizada no Brasil, entretanto, suas construções podem gerar entraves ambientais, entre eles a interrupção da migração de peixes. O presente estudo foi realizado no período anterior a construção da Usina Hidroelétrica (UHE) São Domingos sobre o eixo da cachoeira Branca, no rio Verde -Mato Grosso do Sul (Brazil), e teve como objetivo avaliar a diversidade genética de duas populações naturais de dourado (Salminus brasiliensis) localizadas à jusante (População A -PopA) e montante (População B -PopB) da cachoeira. Foram utilizados oito iniciadores para analisar 56 indivíduos (PopA: 26; PopB: 30). Observaram-se um total de 102 fragmentos, dos quais 86 foram polimórficos (84,3%). Foram identificados fragmentos de baixa frequência (PopA: 2; PopB: 1), três fragmentos limitantes (PopA) e três fragmentos exclusivos (PopB). A variabilidade genética intra-populacional calculada com o índice de Shannon e pela porcentagem de fragmentos polimórficos mostrou altos valores de variabilidade dentro de cada população (PopA: 0,300 e 60,80% e PopB: 0,411 e 79,40%, respectivamente). A distância e identidade genética mostraram uma alta diferenciação genética (0,076 e 0,927, respectivamente). Conclui-se que as populações apresentaram alta variabilidade genética intra-populacional e diferenciação genética entre si, com baixo fluxo gênico.

Most of the electricity used in Brazil comes from hydroelectric plants, mainly due to the great availability of its water resources. However, the construction of these plants denotes serious problems related to migration of native fish and the genetic conservation of stocks. Current study evaluates two wild population of Leporinus elongatus (piapara) located downstream (Population A -PopA) and upstream (Population B -PopB) of the Cachoeira Branca before the construction of the São Domingos hydroelectric plant (HPP) in the Mato Grosso do Sul State, Brazil. Thirty samples from caudal fins were collected and analyzed for each population. Eighty-nine fragments, including 72 polymorphic ones (80.9%), were analyzed. Low fragments (less than 0.100) in both populations (PopA = 2 and PopB = 3) were identified. Nine fixed fragments (frequency 1.000) (PopA = 3 and PopB = 6), and four exclusive fragments (PopA = 3 and PopB = 1) were also reported. The genetic variability within populations, calculated by Shannon Index and by percentage of polymorphic fragments, indicated high rates of intrapopulation variability (PopA = 0.309 and 61.80% and PopB = 0.392 and 71.90%, respectively). Genetic distance and identity rates (0.089 and 0.915, respectively) were different between populations, whilst AMOVA showed that most variations lie within the populations and not between them. Fst and Nm rates showed moderate genetic differentiation with low numbers of migrants. Results reveal populations with high intra-population genetic variability and genetic differentiation, with low gene flow. ladders of São Domingos HPP should control fish transposition to preserve genetic variability. Red‐bellied piranhas (Pygocentrus nattereri) are widely caught with different intensities throughout the region of Solimões‐Amazonas River by local fishermen. Thus, the management of this resource is performed in the absence of any information on its genetic stock. P. nattereri is a voracious predator and widely distributed in the Neotropical region, and it is found in other regions of American continent. However, information about genetic variability and structure of wild populations of red‐bellied piranha is unavailable. Here, we describe the levels of genetic diversity and genetic structure of red‐bellied piranha populations collected at different locations of Solimões‐Amazonas River system. We collected 234 red‐bellied piranhas and analyzed throughout eight microsatellite markers. We identified high genetic diversity within populations, although the populations of lakes ANA, ARA, and MAR have shown some decrease in their genetic variability, indicating overfishing at these communities. Was identified the existence of two biological populations when the analysis was taken altogether at the lakes of Solimões‐Amazonas River system, with significant genetic differentiation between them. The red‐bellied piranha populations presented limited gene flow between two groups of populations, which were explained by geographical distance between these lakes. However, high level of gene flow was observed between the lakes within of the biological populations. We have identified high divergence between the Catalão subpopulation and all other subpopulations. We suggest the creation of sustainable reserve for lakes near the city of Manaus to better manage and protect this species, whose populations suffer from both extractive and sport fishing.
