Genetic Individualization of Domestic Cats Using Feline STR Loci for Forensic Applications

Abstract: A group of ten short tandem repeat (STR) loci suitable for PCR typing from DNA of domestic cats is evaluated for genetic individualization using blinded samples of eight putative feline blood specimens. The ten loci were also typed in a 70 member cat pedigree to demonstrate Mendelian inheritance and independent assortment. A "match window" or measurement precision estimate was empirically established by determining the maximum gel migration difference among alleles identical by descent in different individuals… Show more

“…Of the 17 loci, 14 were unlinked on a genetic recombination map (Menotti- Raymond et al 1999), one was not mapped (FCA166), and one pair was less than 20 cM apart (FCA43 and FCA117). My PCR amplifications followed conditions described by Menotti-Raymond et al (1997).…”

“…The majority of transferred cat hairs have been naturally shed, and lack the large root bulb required for reliable high-discrimination STR profiling [3]. However, even hair shafts typically contain enough mtDNA for sequencing, which is still capable of excluding many potential sources despite lower discriminating power than nuclear DNA [4].…”

“…Their main advantages are that they can be easily assayed by PCR, they are expressed co-dominantly in phenotype, and they are highly polymorphic in most organisms. Recently, there has been an increase in the forensic application of STRs to cases involving non-human DNA, for example, wild animals hunted down by poachers, domestic canines, felines, and even plants [4][5][6][7].…”
