“…6 Another recent report supports the hypothesis that IL-10 protein production is largely under genetic control by showing that heritability in monozygotic twins is 0.75 for the production of IL-10. 7 All these observations emphasise the need for an efficient PCR system, capable of identifying those patients or transplant recipients carrying IL-10 haplo- Table 4 The sequences of the primers used in PCR-SSP method developed to analyse IL-10 promoter polymorphisms. C1 and C2 are control primers specific for exon 3 sequences in HLA-B which were used for HLA phototyping 4 …”

“…To date, however, no single polymorphism (or group of polymorphisms) has been able to explain predisposition toward this phenotype. The importance of this issue was highlighted by the work of Westendorp et al who, in 1997, examined the ex vivo LPS-induced cytokine production capacity of 190 first degree relatives of patients with invasive meningococcal disease [59]. They found that family members of nonsurvivors produced less TNFα and more IL-10 upon ex vivo LPS stimulation than did the relatives of patients who survived, indicating that a predilection towards an anti-inflammatory phenotype may be heritable.…”

“…In an important paper, Westendorp et al 9 showed that the levels of both TNF and IL-10 in firstdegree relatives of patients with meningococcal disease strongly determined the likelihood of patient survival. They also showed a strong concordance of TNF levels in monozygotic twins.…”
