Genetic influence on the associations between IGF-I and glucose metabolism in a cohort of elderly twins

Abstract: There was a high heritability for IGF-I and IGFBP-3, but a low heritability for insulin secretion and insulin sensitivity in a group of elderly twins. In addition, we found a strong negative relationship between IGF-I and insulin sensitivity, which did not seem to be strongly genetically determined.

“…We compared the heritability of serum IGF-I to the heritability of PAPP-A, STC2 and IGF-II, and we used serum IGF-I to assess whether our cohort in an IGF-system context was comparable to other cohorts of elderly subjects. The present heritability index of IGF-I was in agreement with a recent study by Jensen et al, who found a heritability index of 65% using the same IGF-I assay in a different cohort of Danish twins (22). As regards the age relationship and sex difference of serum IGF-I concentrations in elderly people, our results agreed with some previous findings, but not all (25, 53-55).…”

“…To generate such data, we capitalized on our access to a very well characterized cohort of Danish dizygotic (DZ) and monozygotic (MZ) twins (19). To evaluate PAPP-A and STC2 in an IGF context, we also studied the heritability and agedependency of IGF-I and IGF-II (20)(21)(22)(23)(24)(25).…”

“…selected groups of patients. Jensen et al showed a strong negative correlation between IGF-1 and insulin sensitivity, which does not seem to be genetically determined (12). Lower levels of IGF-1 were also confi rmed in patients with non-alcoholic fatty liver disease (13,14).…”
