Genetic Markers of Insulin Resistance and Atherosclerosis in Type 2 Diabetes Mellitus Patients with Coronary Artery Disease

Abstract: Type 2 diabetes mellitus (T2DM) is characterized by impaired insulin secretion on a background of insulin resistance (IR). IR and T2DM are associated with atherosclerotic coronary artery disease (CAD). The mechanisms of IR and atherosclerosis are known to share similar genetic and environmental roots. Endothelial dysfunction (ED) detected at the earliest stages of IR might be the origin of atherosclerosis progression. ED influences the secretion of pro-inflammatory cytokines and their encoding genes. The genes… Show more

“…Regarding MetS-related MCs activation (51), the aforementioned mediators contribute to the onset of IR by compromising the insulin signaling pathways (51), which is associated with MetS-related atherosclerosis (128). The relationship between atherosclerosis and MetS is multifactorial, with obesity, dyslipidemia, and IR playing substantial roles in both its onset and progression (129).…”

“…MetS is closely linked with MC activation [135]. The mediators released by MCs contribute to IR, thereby promoting the development of atherosclerosis [135], a condition strongly associated with MetS [136]. The correlation among atherosclerosis and MetS is multifactorial, involving MetS-related parameters like IR as substantial contributors to its onset and progression [137].…”

“…All its stages from atheroma formation and growth to complications (e.g., myocardial infarction [2] and stroke [3] as the leading causes of death, according to the World Health Organization (WHO) [4]) represent the inflammatory response to the lesion caused by specific cytokines. Atherosclerosis is an aging-related disease [5], with such factors as acquired dyslipidemia [6], acquired diabetes [7], tobacco smoking [8], past diseases [9], lack of physical activity and excessive physical activity that are collectively called lifestyle increasing the risk to develop it. However, atheromas in artery walls can be detected pathoanatomically even as early as during embryogenesis [10].…”
