Genetic polymorphisms in MMP 2, 9 and 3genes modify lung cancer risk and survival

Abstract: BackgroundMatrix metalloproteases (MMPs) are proteolytic enzymes that contribute to all stages of tumour progression, including the later stages of invasion and metastasis. Genetic variants in the MMP genes may influence the biological function of these enzymes and change their role in carcinogenesis and progression. We have investigated the association between the -735 C/T, the -1171 5A/6A, and the -1562 C/T polymorphisms in the MMP2, MMP3 and MMP9 genes, respectively, and the risk and survival of lung cancer… Show more

“…Many studies have investigated the role of MMP-2 polymorphisms in lung cancer risk, but have yielded inconsistent and inconclusive results (Yu et al, 2002;Zhou et al, 2005;Rollin et al, 2007;Song et al, 2007;Aysegul et al, 2011;Gonzalez-Arriaga et al, 2012). Meta-analysis is a quantitative approach in which individual study findings on the same topic are statistically integrated and analyzed, and recently, it has been used for the evaluation of cancer prognostic markers and genetic risk factors.…”

“…After discarding those which clearly did not meet the criteria and excluding 19 records, 7 full-text publications with a total of 3,189 lung cancer cases and 3,013 controls were included into this meta-analysis (Yu et al, 2002;Zhou et al, 2005;Rollin et al, 2007;Song et al, 2007;Jia, 2009;Aysegul et al, 2011;Gonzalez-Arriaga et al, 2012). Ethnic groups among these studies were as following: 3 from Caucasians and 4 from Asians.…”

“…42 The enzymatic activity and expression of MMP-2 were examined using gelatin zymography and western blotting assays, respectively ( Figure 6). When both cell lines were treated with either SQE or CIS, lower levels of MMP-2 enzymatic activity were detected, along with reduced levels of MMP-2 expression.…”

“…Matrix metalloproteinases (MMPs) are a family of zinc-dependent protease enzymes that degrade and restructure the extracellular matrix (ECM) components and basal lamina which are physical barriers for cancer cells [6,7,8]. MMPs have a broad substrate activity including the ECM components of collagen type I-VIII, gelatin, elastin, laminin, and also myelin basic protein and several growth factors.…”
