Growing evidence suggests that the transforming growth factor b (TGF-b) signaling pathway occupies a central position in the signaling networks that control cell growth and differentiation. TGFb1 and its receptor TGF-bRII have been correlated with the development of certain forms of cancer, including gastric cancer. We hypothesized that functional genetic variants in TGFB1 and TGFBR2 are associated with gastric cancer risk. To test this hypothesis, we genotyped C-509T and Leu10Pro polymorphisms in TGFB1 and G-875A variant in TGFBR2, using the primer-introduced restriction analysis (PIRA)-PCR assay, in a case-control study of 675 gastric cancer cases and 704 healthy controls in a Chinese population. We found that the variant alleles of the promoter polymorphisms, TGFB1 C-509T and TGFBR2 G-875A, were associated with a significantly decreased risk of gastric cancer [adjusted odds ratio (OR) 5 0.65, 95% confidence interval (CI) 5 0.52-0.82 for 2509CT/TT and adjusted OR 5 0.67, 95% CI 5 0.53-0.85 for 2875GA/AA]. Furthermore, subjects with both variant genotypes of the TGFB1 C-509T and TGFBR2 G-875A were associated with a significantly (56%) decreased risk of gastric cancer (adjusted OR 5 0.44, 95% CI 5 0.32-0.62). These findings indicate, for the first-time, that the functional variants in the promoter of TGFB1 and TGFBR2 might contribute to gastric cancer susceptibility. ' 2006 Wiley-Liss, Inc.

BACKGROUNDIt has been suggested that genetic polymorphisms in certain glutathione‐S‐transferase (GST) genes reduce the effectiveness of detoxifying cytotoxins generated by chemotherapeutic agents, potentially resulting in enhanced clinical responses to chemotherapy.METHODSThe authors evaluated common polymorphisms in the GSTM1, GSTT1, and GSTP1 genes for associations with overall survival in 1034 patients with invasive breast carcinoma who were recruited into the Shanghai Breast Cancer Study between 1996 and 1998, treated with chemotherapy, and followed for a median of 5.3 years.RESULTSAfter adjusting for age, tumor stage, and the use of radiotherapy and tamoxifen, women who were homozygous for the variant GSTP1 105Val allele had a 60% reduction in mortality risk compared with women who were homozygous for the Ile allele (hazard ratio, 0.4; 95% confidence interval, 0.2–0.8). No association was found with respect to any of the GSTM1 or GSTT1 genotypes.CONCLUSIONSThe results of the current study indicate a potential role for GSTP1 polymorphism in predicting the clinical outcomes of patients with breast carcinoma who are treated with chemotherapy. BACKGROUND.The ataxia‐telangiectasia mutated (ATM) gene plays a critical role in cell‐cycle arrest, apoptosis, and DNA repair. However, to date, no study has directly investigated the association between ATM gene expression and breast cancer survival.METHODS.ATM gene expression levels were evaluated in tumor and adjacent normal tissue from patients diagnosed with primary breast cancer or BBD using quantitative real‐time reverse transcription polymerase chain reaction (RT‐PCR) assays. Cox regression models were used to evaluate the association of ATM gene expression and survival in a cohort of 471 breast cancer patients.RESULTS.In breast cancer cases, ATM expression in cancer tissues was decreased by approximately 50% compared with adjacent normal tissues from the same patients. In BBD cases, the expression level of the ATM gene was similar in benign tumor tissue and adjacent normal tissues. No apparent difference was found in ATM gene expression levels in adjacent normal tissues obtained from cancer patients or BBD controls. Compared with patients with the lowest tertile of the ATM mRNA, patients in the upper 2 tertiles had more favorable disease‐free survival (hazard ratio [HR] = 0.46, 95% confidence interval [CI]: 0.30–0.73 and HR = 0.52, 95% CI: 0.33–0.81, respectively) and overall survival (HR = 0.56, 95% CI: 0.35–0.92 and HR = 0.70, 95% CI: 0.43–1.13, respectively).CONCLUSIONS.The ATM gene expression was down‐regulated in breast cancer tissues and a high ATM gene expression level in breast cancer tissue was associated with a favorable prognosis.
