Genetic Relationship amongSome Wheat Genotypes Using Ten ISSR Markers

Abstract: Ten of ISSRs (inter-Simple Sequence Repeats) markers were used for revealing genetic relationship, genetic diversity and DNA fingerprint of ten bread wheat (Triticumaestivum L.) genotypes. Primers varied among them in giving unique DNA fingerprint hence primers HB12, UBC811andUBC852 gave unique fingerprint for all ten studied wheat genotypes, others varied between 2-6 fingerprint genotypes while primer 17899A gave no unique fingerprint. High genetic distance was 0.661 while low genetic distance was 0.131.Clust… Show more

“…The highest number of primary and polymorphic bands was 18 and 10, respectively, in primer OPA-10. Recognition of the highest number of annealing sites by primer usually results in a high number of main bands, also established and reported in past studies in maize (Al-Tamimi and Al-Saadi 2018), in wheat (Tahir, 2014;Al-Ghufaili, 2017), and that later enhances the chances to provide polymorphic bands.…”

“…Among DNA markers, ISSR and RAPD require small quantities of DNA and do not require radioactive labels; they are simple and fast. These markers are reproducible, powerful, simple, quick, and relatively cheap for use in estimating genetic variation and identifying differences between populations [11,13,28]. In the present study, both RAPD and ISSR markers generated reproducible and distinct banding patterns.…”

“…,since all treatments belong to coriander , appearance related to this relationship (Al-Judy.,2004) because genome contains conserved sequence retained in all treatments including those arises from irradiation treatments) ,these shared sequences appeared in form of monomorphic bands (Russel et al, 1997;,AL-Badeiry, 2013 andAl-Tamimi,2020). Primer OPA-10 gave highest number unique bands ( reached to seven bands ) ,primer ability of to recognize a unique annealing site on genome success in producing a unique DNA fingerprint for a particular genotype (Fadoul et al,2013 ;AL-Ghufaili, 2017, AL-Saadi.,2018,AL-Tamimi.,2021. Primer OPC-19 gave highest value for amplified bands , primer OPA-10 gave highest value for main bands ,these two criteria concerned primer ability to recognize more annealing sites.…”
