Genetic screening and mutation identification in a rare canine breed, the Drentsche patrijshond

Abstract: The Drentsche patrijshond (Drent) is a relatively rare breed that is used as a versatile hunting dog. Although inherited diseases occur in this breed, no genetic mutations were previously known to contribute to these inherited disorders. Thirteen Drents were screened for 142 known disease-associated mutations that occur in domestic dogs. Of these, two mutations were identified to segregate in three pedigrees: mutations for hyperuricosuria and von Willebrand disease type 1. This information can be used to scree… Show more

“…Samples were obtained primarily from buccal swabs, whole blood, and docked tail tissue. DNA was extracted using routine, standard methods as previously described [Shaffer et al, 2015[Shaffer et al, , 2016[Shaffer et al, , 2017 24 PMEL SINE Insertion Size Analysis PCR amplification of the region flanking the SINE insertion site in PMEL was performed using 2 different primer sets to verify and confirm the results. The primer sequences for the 2 assays are: * VIC * F-5 ′ -CCATAGATAGGCTTTCCTTGTCC, R-5 ′ -GGGCA-AGAGCGGAAGACC and * FAM * F-5 ′ -CAGTTTCTCCTTTA-TTCTCCCAATG and R-5 ′ -GAGAGCTGAGCCCTGCTTC.…”

“…Samples were then collected on 13 additional CF dogs specifically to identify carriers or at-risk dogs at the superoxide dismutase 1 gene ( SOD1) locus. For 22 dogs, three buccal swabs per dog were used for sample collection and DNA was extracted using the 5Prime buccal brush protocol as previously described (Shaffer and others 2015). Frozen semen samples were used for DNA extraction for six dogs; whole peripheral blood was used for two dogs.…”

“…The mutations analysed have been reported previously (Shaffer and others 2015), with the following exceptions: Table 1 shows the disease tests performed on the drents (Shaffer and others 2015) but not used in the current study and Table 2 shows 14 additional mutations studied in the CF dogs but not in the drents.…”

“…Other breeds included Bloodhound (6), English Springer Spaniel (1), Australian Shepherd (1), Belgian Sheepdog (2), Border Collie (3), Catahoula Leopard Dog (2), Czechoslovakian Vlcak (1), Golden Retriever (4), Standard Poodle (1), and mixed-breed (14) 200 DNA was extracted using routine standard methods. Mutation regions were investigated using a variety of methods depending on the specific mutation, as previously described [Shaffer et al, 2015[Shaffer et al, , 2016. Results of testing were provided to handlers or superior officers upon request.…”
