Genetic variability in Metarhizium flavoviride revealed by telomeric fingerprinting

Abstract: Previous studies using arbitrarily primed PCR (AP-PCR/RAPD) analysis have shown only little genetic variation among isolates of the entomopathogenic fungus Metarhizium flavoviride. In the current study, however, telomeric fingerprinting unambiguously differentiated several Brazilian strains of M. flavoviride as well as strains from Africa and Australia. Using this technique, similarity estimates of telomeric DNA among distinct strains were less than 50%, showing this locus to be highly mutable in this species.… Show more

“…Telomeric probes have previously been used for DNA fingerprinting of filamentous fungi such as U. maydis (Sanchez-Alonso et al 1996), Metarhizium sp. (Inglis et al 1999), Nomuraea rileyi (Boucias et al 2000), Cercospora caricis (Inglis et al 2001b) and Beauveria sp. (Viaud et al 1998).…”

“…Extraction of fungal genomic DNA from liquid-grown mycelium, and analysis of telomeric RFLPs were described previously (Inglis et al 1999). In order to map the extent of the element associated with the monomorphic telomeres in strain CG34, genomic DNA was digested with a range of restriction enzymes, and Southern blots were probed with the telomeric sequence from Fusarium oxysporum [TTAGGG] 18 (Powell and Kistler 1990).…”

“…Intra-specific genetic polymorphism in entomogenous fungi has facilitated strain characterization often employing a range of techniques (e.g. St Leger et al 1992;Pfeifer and Khachatourians 1993;Viaud et al 1996;Couteaudier and Viaud 1997;Maurer et al 1997;Inglis et al 1999;Enkerli et al 2001;Coates et al 2002a,b). Most often the markers can distinguish at the species but not at the strain level.…”
