Genetic variation at the catalytic subunit of glutamate cysteine ligase contributes to the susceptibility to sporadic colorectal cancer: a pilot study

Abstract: Background Glutathione is a tripeptide detoxifying a variety of exogenous and endogenous free radicals and carcinogens, and a deficiency of glutathione is associated with an increased host susceptibility to oxidative stress, a pathological condition implicated in the development and progression of cancer. The catalytic subunit of glutamate-cysteine ligase (GCLC) is an enzyme responsible for the initial and rate-limiting step of glutathione biosynthesis. Methods and resultsThe aim of this pilot study was to inv… Show more

“…Additionally, a significant genotype × gender interaction was found in the rs79361899 recessive model. Gender-specific associations between certain polymorphisms and various types of cancer have frequently been observed, including gallbladder carcinoma [ 21 ], lung cancer [ 2 ], and colorectal cancer [ 22 ]. The exploration of SNP interactions facilitates understanding the intricate gene-environment interplay (G × E) [ 23 ].…”

“…Genetic polymorphisms of glutamate cysteine ligase that are correlated with a decrease in GCLC mRNA and protein expression, enzyme activity, and GSH content [ 47 , 48 , 49 , 50 , 51 ] represent attractive markers for studying the molecular mechanisms of psoriasis. Polymorphisms of the GCLC gene have been found to be associated with the risk of cardiometabolic diseases such as coronary artery disease [ 52 , 53 ], ischemic stroke [ 54 ], type 1 [ 55 , 56 ] and type 2 [ 51 ] diabetes mellitus, polycystic ovary syndrome [ 57 ], and nonalcoholic fatty liver disease [ 58 ], as well as other multifactorial disorders such as bronchial asthma [ 59 ], pulmonary tuberculosis [ 60 ], and colorectal cancer [ 61 ]. However, no studies have been designed so far to investigate the role of GCLC gene polymorphisms in psoriasis susceptibility.…”

“…The activation of NRF2 can regulate the transcription of ARE with cytoprotection and detoxification effects, such as HO-1 and NQO1, to maintain cell redox homeostasis and reduce cell oxidative stress [58]. The catalytic subunit of GCLC is an enzyme that plays a pivotal role in initiating and controlling the rate-limiting step of glutathione biosynthesis, and the lack of glutathione is related to the increase in the sensitivity of the host to oxidative stress [59]. Following ROS exposure, NRF2 migrates to the cell The stimulation of H 2 O 2 can cause the oxidation of protein as well as DNA damage in cells and constantly produces •OH radicals, thus leading to the generation of new H 2 O 2 and triggering a chain reaction of oxidative stimulation [54].…”
