Genetic Variation of Butternut Squash (<i>Cucurbita moschata</i> Duchesne) based on Inter-Simple Sequence Repeat

Abstract: Butternut squash (Cucurbita moschata) is a Cucurbitaceae plant that has been widely cultivated in Indonesia. Butternut squash is known to have various cultivars. A new cultivar introduced by the Faculty of Biology UGM is named ‘Citra Laga’ which is expected to be able to compete with the imported cultivars. The number of cultivars within a species may indicate genetic variation. This research was conducted to observe genetic variation and the phenetic relationship between ‘Citra Laga’ and the imported butternu… Show more

“…The similarity index may indicate their genetic relationship. If the similarity value is high, then the genetic relationship between them will also be close (Putri et al 2023). The results shows that the analysis of phenetic relationship between P. acidula at the three locations and S. alba from Biluhu yielded in 3 clusters, namely A, B and C. Cluster A consist of P. acidula at Biluhu and Dulanga with similarity of 100%.…”

“…The DNA band patterns produced by ISSR markers were converted into a binary matrix 0-1 based on the absence or the presence of the DNA band (Ibrahim 2021). They were analysed using MVSP version 3.1 program with simple matching coefficient of UPGMA method to construct dendogram based on similarity index (Singh et al 2014;Putri et al 2023).…”

“…Recent studies conducted by Putri et al (2023) using ISSR markers employed several cultivars of C. moschata and found that only 18.6% polymorphism rate was recorded. The variability of pumpkin genetics can be partly explained by their biotic and abiotic environmental factors.…”

“…The use of plant isozyme data for the identification and classification of plants has been employed in Ranunculus nanus (Suranto 2002), Cassia auriculata (Siva and Khrisnamurthy 2005), Oryza sativa (Widiyanti et al 2008), almond (Colic et al 2010), Amorphophallus (Anil et al 2014), and C. moschata (Jiang et al 2023). Accordingly, studies of C. moschata using genetic markers were conducted by Lee et al (2021) and Putri et al (2023) testing their genetic structure and genetic diversity respectively.…”
