Genetic Variation of Yellow Barbell (Carasobarbus Luteus (Heckel, 1843)) From Four Populations Using Mitochondrial Dna Coi Gene Sequences

Abstract: The aim of this study is to determine genetic variation of C. luteus populations by conducting gene sequence analysis of mtDNA COI locus. Total DNA of 48 individuals from 4 populations in total was isolated using a commercial kit. Sequence analysis was conducted through 3500 XL Genetic Analyzer device following the amplification of target mtDNA locus via Polymerase Chain Reaction (PCR). With sequence analysis of mtDNA COI locus, 9 polymorphic sites and 4 haplotypes were determined. Haplotype and nucleotide div… Show more

“…Furthermore, this possibility was supported by low nucleotide diversity as well. Another study on mtDNA COI sequence analysis of C. luteus species in the same river system revealed a considerable divergence from neutrality (Parmaksız & Eskici, 2018). However, it was expected that genetic data resulted in greater details because the number of samples and localities were higher in this study.…”

“…In addition, it was determined that either of mean Tajima's D and Fu's Fs values were negative in the study by Parmaksız and Eksi (2017), on the contrary, both of them were positive in the study by Parmaksız and Eskici (2018). A positive Fu's Fs and Tajima's D test signifies lower levels for both low and high frequency polymorphisms, indicating a decrease in population size (Hu et al, 2016 ).…”

“…The primer sequence used for amplification of the mtDNA COI gene region was reported by Darabi et al (2014) (COI-625F: 5' TCA ACC AAC CAC AAA GAC ATT GGC AC-3'; COI-625R: 5' GAC TTC TGG GTG GCC AAA GAA TCA-3') [19]. PCR conditions and chemicals were applied according to the study of Parmaksız and Eskici (2018) and the product was obtained [20]. The primer sequence used for the mtDNA cyt b gene region was taken from Briolay et al, (1998) (L15267 F: 5ˈ GTT TGA TCC CGT TTC GTG TA-3ˈ; H15891 R: 5'AAT GAC TTG AAG AAC CAC CGT-3'), PCR conditions and chemicals were applied according to the study of Parmaksız and Şeker (2018) and the product was obtained [21,22].…”
