Genome Analysis of a Novel Bradyrhizobium sp. DOA9 Carrying a Symbiotic Plasmid

Abstract: Bradyrhizobium sp. DOA9 isolated from the legume Aeschynomene americana exhibited a broad host range and divergent nodulation (nod) genes compared with other members of the Bradyrhizobiaceae. Genome analysis of DOA9 revealed that its genome comprised a single chromosome of 7.1 Mbp and a plasmid of 0.7 Mbp. The chromosome showed highest similarity with that of the nod gene-harboring soybean symbiont B. japonicum USDA110, whereas the plasmid showed highest similarity with pBBta01 of the nod gene-lacking photosyn… Show more

“…Compaction of sym genes arrangement has been indicated in primary rhizobia. While the majority of Bradyrhizobium strains harbour these genes in several non-linked chromosomal loci, in some strains these genes are concentrated on symbiotic plasmids (pSyms) [12]. A more compact arrangement was found in Mesorhizobium species, which presumably originated from sym gene transfer into Phyllobacteriaceae strains [7].…”

“…DOA9 is also original among bradyrhizobia. It is the only Bradyrhizobium strain in which the classical symbiotic genes (nod, nif, fix and T3SS) are harboured on a mega plasmid (named pDOA9), in the other Bradyrhizobium strains, these genes are generally clustered on a genomic island (Okazaki et al, 2015). This strain, which is phylogenetically distant to the photosynthetic bradyrhizobia, was isolated from a paddy field using A. americana as a trap plant (Noisangiam et al, 2012).…”
