Genome Sequences of Bacillus sporothermodurans Strains Isolated from Ultra-High-Temperature Milk

Abstract: Here, we report the draft genome sequences of 3 Bacillus sporothermodurans strains isolated from ultra-high-temperature milk products in South Africa and Brazil and the type strain MB 581 (DSM 10599). The genomes will provide valuable information on the molecular dynamics of heat resistance in B.

“…Bacillus sporothermodurans PL01 is a bacterium which produces highly temperature-resistant endospores [48]. In contrast to green algae C. vulgaris, the photometer signal of PL01 increases from a cell density of 2•10 7 mL −1 on, whereas the autofluorescence intensity can be measured beginning from a cell density of 2•10 8 mL −1 (Figure 10D,H).…”

“…Bacillus sporothermodurans strains SAD, SA01, BR12, and DSM 10599, as well as B. oleronius DSM 9356, were sequenced and annotated as previously described [15,16]. Subsequent analyses in this study are based on the strains mentioned above.…”

“…Differences in heat resistance are thought to be clonal, which explains the growing interest in the identification of evolutionary associations and the understanding of its high heat resistance. In this study, the whole genomes of B. sporothermodurans strains isolated from UHT milk from South Africa and Brazil, the type strain, and B. oleronius , its closest phylogenetic neighbor, were sequenced [ 14 , 15 ], with the aim of comparing proteins involved in heat resistance between B . sporothermodurans and other Bacillus species to better understand this phenotype in B .…”

“…Circular chromosomes were rotated to dnak as starting point. Possible contamination of the assemblies was screened using the ContEst16S tool [ 22 ]. The genome quality was assessed with the PATRIC quality tool [ 21 ].…”

“…It currently provides the best possibility to clarify genomic relationships with the highest discriminatory power [ 21 ]. At the time of writing, the genome of the H. vini type strain was not published and the genome sequences of only five H. sporothermodurans strains were available, i.e., those of the type strain DSM 10599 T , strain BR3 (also named BR12), strain SAD, strain SA01 and B4102 [ 15 , 22 ]. Their assemblies consist of 100–800 contigs, which is a relatively high number when compared to other bacterial species.…”
