Genome-Wide Analysis of DNA Methylation before-and after Exercise in the Thoroughbred Horse with MeDIP-Seq

Abstract: Athletic performance is an important criteria used for the selection of superior horses. However, little is known about exercise-related epigenetic processes in the horse. DNA methylation is a key mechanism for regulating gene expression in response to environmental changes. We carried out comparative genomic analysis of genome-wide DNA methylation profiles in the blood samples of two different thoroughbred horses before and after exercise by methylated-DNA immunoprecipitation sequencing (MeDIP-Seq). Different… Show more

“…As shown in Supporting information Figure S3 , the frequency of methylation (7%) was significantly higher at promoters than downstream of transcription start sites (≤3 kb) in each group. In contrast, gene-body regions (intergenic, intron, exon regions) exhibited higher frequency of methylation, which was in agreement with previous studies 32 , 34 . However, the most significant effects on gene expression were associated with altered DNA methylation in the promoter regions, which frequently contain CpG sites 30 .…”

“…It is believed that DNA methylation regulates many biological processes, including gene expression, genomic imprinting, and X chromosome inactivation [ 5 7 ]. Using the methylated DNA immunoprecipitation-sequencing (MeDIP-seq) technology, methylation studies have been extensively conducted in animals [ 8 10 ].…”

“…Regions were considered significantly methylated if the edgeR.p-value was below 0.01 and if the number of reads, of a specific region, in one of the samples was higher than the mean of reads of all regions (the whole genome), which is the background correction. This p-value was derived from other studies performing MeDIP-seq analysis [9] , [10] , [11] .…”
