Genome-Wide Analysis of the Binding of the Hox Protein Ultrabithorax and the Hox Cofactor Homothorax in Drosophila

Abstract: Hox genes encode a family of transcription factors that are key developmental regulators with a highly conserved role in specifying segmental diversity along the metazoan body axis. Although they have been shown to regulate a wide variety of downstream processes, direct transcriptional targets have been difficult to identify and this has been a major obstacle to our understanding of Hox gene function. We report the identification of genome-wide binding sites for the Hox protein Ultrabithorax (Ubx) using a YFP-… Show more

“…have reported 570 genes as putative targets of Ubx in Drosophila , identified by ChIP-chip analysis of Cbx wing discs using anti-Ubx antibodies specific to the N-terminal region of the protein. 219 of these targets were common to those reported by Choo et al 8. (Suppl.…”

“…35,36 In addition, whole genome ChIP-chip analyses of Dorsal, Twist, and Snail in Drosophila , have also shown similar results, 37,38 in which these TFs appeared to exert positive/negative regulation of hundreds to thousands of target genes for dorsoventral patterning, anteroposterior patterning, and mesoderm differentiation. Our ChIP-seq analyses for Otx2, Lim1, and Gsc in early vertebrate embryos are consistent with work in Drosophila .…”
