Genomic characterization of six novel Bacillus pumilus bacteriophages

Abstract: Twenty-eight bacteriophages infecting the local host Bacillus pumilus BL-8 were isolated, purified, and characterized. Nine genomes were sequenced, of which six were annotated and are the first of this host submitted to the public record. The 28 phages were divided into two groups by sequence and morphological similarity, yielding 27 cluster BpA phages and 1 cluster BpB phage, which is a BL-8 prophage. Most of the BpA phages have a host range restricted to distantly related strains, B. pumilus and B. simplex, … Show more

“…The contributions of the SEA-PHAGES students have been essential to our current understanding of the diversity of mycobacteriophages, demonstrating the substantial impact of the distributed approach compared to what would be accomplished by a single laboratory, and have resulted in several publications with student authors (29, 3139). Since the start of the program in 2008, SEA-PHAGES students have isolated 3,000 new phages (with global positioning system [GPS] coordinates recorded) and characterized their phages by DNA restriction analysis and electron microscopy.…”

“…Each of the two phages had an isometric head with a diameter of 88 nm, a 189-nm-long contractile tail, a 60-nm-long tail needle, and a 20-nmlong and 46-nm-wide baseplate. To date, a few phages of B. pumilus have been isolated (17), while only one B. pumilus phage from the Myoviridae family has been morphologically characterized (16).…”

“…Before 1981, more than 33 different phages were reported to infect B. pumilus, and a few more have been characterized since (15). In addition, the genome of 10 B. pumilus phages have been characterized to date (16,17); however, the structural proteome of the phage virion has not yet been analyzed.…”

“…Phamerator is popular among the large groups studying Mycobacteriophages [4] and Bacillus phages [5, 6] and is steadily gaining traction in other areas of phage research [711]. …”
