Genomic insights into a robust gamma-aminobutyric acid-producer Lactobacillus brevis CD0817

Abstract: Lactobacillus brevis CD0817, a strain isolated from a healthy adult gut, was currently the most efficient lactic acid bacterial cell factory for gamma-aminobutyric acid. In this study, the complete genome sequence of CD0817 was determined and compared with some related L. brevis genomes. The CD0817 genome consists of one 2,990,570-bp chromosome and four plasmids. The comparative genomic and phylogenetic analysis revealed that L. brevis CD0817 was not ver… Show more

“…brevis strains, with only 91% of amino acid sequence identity. The results further demonstrated that GadB contributed to the main GAD activity [55]. gadA does not have a close relationship with the accumulation of GABA.…”

“…Recently, it was found that a high GABA-producing Lb. brevis CD0817 contained only a gadB-gadC operon and was absent of gadA gene [55]. What is more, there are obvious differences between GadB of CD0817 and those in the other GABA-producing Lb.…”

“…Typically, L. brevis contains two GAD-encoding genes, gadA and gadB, which when expressed yield GAD enzymes that share approximately 50% amino acid sequence similarity [6]. In contrast, the gadB gene is absent in strain L. brevis CD0817 [59] and the amino acid sequence identities of GadA and GadC from L. brevis CD0817 against other L. brevis strains are 91% and 90%, respectively. The transcriptional regulator gene gadR plays a crucial role in GABA production and acid resistance in L. brevis.…”

“…Microorganisms, including LAB, are an important source of GABA [ 1 , 39 42 ], and the GABA producing capability amongst LAB like lactobacilli varies between species and strains [ 43 – 47 ]. More recently, high GABA producing lactobacilli strains have been isolated to meet consumer needs [ 45 , 48 ]. In this manuscript, we sought to further investigate strain-specific mechanisms for GABA production in Levilactobacillus brevis .…”
